Treatment of Multiple Myeloma: Something Old, Something New

Multiple Myeloma is an incurable plasma cell malignancy characterised by anaemia, bone destruction and a monoclonal protein in the plasma and/or urine. Recent research has been directed at the mechanisms of the interaction between the neoplastic clone and the bone marrow microenvironment. This has led to trials of new agents for induction as well as for relapsed or refractory disease. Results indicate that immunomodulatory drugs may provide convenient alternatives to melphalan/prednisolone as first-line therapy. Proteasome inhibitors also show promise in combination with dexamethasone. Biphosphonates reduce the skeletal complications of the disease, and may contribute to apoptosis of the malignant clone.....
